Now I have so many ideas for books where obviously there is a crime in them, but that is not the point of the story … For me a book is always about the emotional journey. The plot is completely irrelevant as long as you can take people on that journey with you, say to them: ‘You know what? Terrible things are going to happen but if you trust me you’ll be emotionally satisfied.’ That’s why I’m not even interested in solving the crime, sometimes I don’t even care if you know what exactly happens or whodunnit. In addition, we studied cell permeability of [ 18F]FTBG by confocal imaging of SNAP-tag + cells. Cells were either pre-dosed with [ 18F]FTBG for 30 min or left untreated followed by staining with the cell-impermeable SNAP-tag dye BG-Dy549P1and the highly cell-permeable dye SNAP-Cell® 647-SiR. Pre-dosing of cells with [ 18F]FTBG largely abolished fluorescent labeling of both extra- and intracellular epitopes ( Fig. 2D) while we observed bright staining of these structures in untreated SNAP-tag + cells ( Fig. 2C). This together with the above radiotracer uptake experiments suggests that [ 18F]FTBG is cell-permeable.

Even though foreign gene finders may perform sub-optimally, their predictions may display compositional properties of the novel genome. For example, when annotating the A. thaliana genome with a C. elegans gene finder, the predictions appeared very much like real A. thaliana genes. Figure 3d shows a splice acceptor pictogram derived from these predictions. Note that the sequence composition broadly resembles true A. thaliana splice acceptors, including a preference for G at -3 and a T-rich upstream sequence. It also retains some C. elegans qualities such as a greater proportion of Ts at -5 and -6. When new genomic sequence emerges, it must be annotated with something, and frequently this is a gene finder for a completely different genome. A particularly good example of this is Genscan [ 1], which was trained primarily for the human genome, but has been used to annotate genes in worms, flies, fish, fungi, amphioxus, and others [ 2– 6]. Since sequence features such as codon bias [ 7] and splicing signals [ 8] vary from organism to organism, it is expected that gene finders may not perform optimally in a foreign genome. While the practice of annotating a genome with a foreign gene finder is commonplace, its consequences are not widely understood. Luca Anzilotti and Michael Münzing started working together in 1985 in the group Off (Organisation For Fun), with Sven Väth. They recorded two albums, Organisation for Fun (1988) and Ask Yourself (1989), and a series of singles, including " Electrica Salsa", until 1990. [ citation needed]
